Despite the reported and conceived smalland local impacts at small and medium-sizedoffshore wind farms, the experiencewith future large-scale wind farms mayshow otherwise. At the same time theindustry now faces daunting logistic and scientific challenges as the constructionsites move offshore both in relation to theassessment of the status of habitats andspecies, and in relation to the estimationof environmental effects.
\n\t\t\tThe key problems are lack of reliable models both of the distributional dynamics and of the habitat displacement and related impacts on populations of the species in question. This situation has hampered decision-making in relation to the management of the offshore wind energy sector by introducing unnecessary conflicts with conservation interests. As shown in this paper habitat models may offer solutions to many environmental barriers by providing data in high spatio-temporal resolution about the distribution of sensitive species.
\n\t\t\tDetailed data about the distribution of sensitive species is required in order to:
\n\t\t\tPredict likely changes in distribution arising from natural dynamic change in the marine environment;
Evaluate more accurately the potential loss of habitat arising from exclusion (displacement) of priority and sensitive fauna from offshore wind farm areas as induced by disturbance and underwater noise emissions;
Assess the impact of cumulative habitat loss on priority and sensitive species arising from wind farm construction;
Avoid conflicts in future offshore wind energy schemes associated with environmentally sensitive areas.
The programmes of biological sampling that are typically carried out for the offshore industry have documented problems associated with biological sampling in a dynamic environment. Even benthic habitats are not stable, and as the weather windows during which sampling of species and habitats is typically undertaken are relatively small interpretation and generalisation of results from baseline surveys is often constrained. Examples of such constraints are the lack of information on the distribution of food supply to higher trophic levels like birds, and the lack of information on the variation of habitats at the site. Thus, the next generation of habitat models does not only require inclusion of dynamic variables, but also requires the application of a process-based approach which integrates ecosystem models and statistical models.
\n\t\t\tThis paper highlights some examples of integrated, dynamic ecosystem and habitat models, which have been applied in relation to recent offshore wind farm projects in Denmark. Time will tell whether dynamic, process-driven habitat models will form the benchmark for future impact assessments in offshore areas, and whether developers and regulators will have access to solid descriptions of local environmental conditions with lower risks for the appearance of unforeseen impacts and environmental barriers (ON/OFF News, 2010).
\n\t\tIntegrated models can enable offshorewind farm projects to better demonstrateecological sustainability in offshorewaters, even in the presence of tight timeschedules for baseline investigations. Due to the variability of environmentaleffect parameters in dynamic offshoreenvironments, the risk exists that majordynamics and changes remain undetectedby traditional measurements and monitoring,even following prolonged andintensive sampling campaigns. In mostcases, developers will be requested toprovide solid descriptions of the environmentalbaseline conditions based on investigationscarried out over a relativelylimited period of time. Thus, results of baseline investigations in offshore environments are often constrained due to the following factors:
\n\t\t\tUneven coverage;
Short weather windows;
Short baseline period;
This situation may have pronounced financial consequences and may give rise to speculations on the scale of possible effects. The experience from the most recent constructions of offshore wind farms shows that the time schedules under which baseline investigations have to be undertaken will be very tight. In some countries like Germany two years of baseline sureys is mandatory, however in other countries like Denmark baseline studies related to the last large-scale projects (Horns Rev 2, Rødsand 2 and Anholt) were undertaken over just one year. Ecological conditions for many offshore sites on the basis of one year of investigations may not be sufficient to detect major dynamics, and may lead to flawed conclusions on the presence of priority habitats and species at or near the site.
\n\t\tOptimization of habitat models in the marine environment requires the development of models which are both sufficiently detailed to describe the realized niche occupied by the species in focus and at the same time sufficiently generalized and parsimonious to be able to predict distributions for a range of environmental scenarios. In other words the next generation of marine habitat models needs to include predictor variables which reflect the whole range of scale-dependent processes which form the basis for the distribution of the species at the site. Since marine processes are dynamic by nature marine habitat models need to be designed in a way which describes the range of dynamics of the key processes driving the distribution of the species.
\n\t\t\tIn the environmental programmes related to two of the latest large-scale offshore wind farm project; Horns Rev 2 (2008-2010) and Anholt(2009-2010) DHI used the MIKE modelling framework (Rasmussen, 1991) to facilitateeasy and seamless linking of all modelsrequired for the full implementation of alocal model in relation to the various aspectsof the feasibility, construction andoperation of the wind farm. The MIKEmodelling framework links the basichydrodynamic and wave modules to thedifferent modules applied for sedimentationprocesses, water quality, and benthicpelagic environmental conditions.The water level variation and flows are simulated in response to a variety of forcing functions using a stratified model, MIKE3 (DHI Water & Environment, 2000). The water levels and flows are resolved on an arrayof nested regular grids.
\n\t\t\tBenthic habitat models have been developed reflecting thelinks between the variability of thelong-lived elements and bio-coenoces ofbenthic communities in the regions surrounding the sites in the central Kattegat and the North Sea, and measured/modelled parameterslike water depth, sediment, sedimentgrain size, water temperature, oxygenlevel, contents of organic matter, lightattenuation, plankton density, density ofsuspended material in the water columnetc.The resulting statistical species distribution models are directly coupled to the refined hydrodynamic modelswhich produced temporally resolvedpredictions of local distribution changes of benthic fauna and flora resulting from natural changes in oceanographicconditions.The statistical models could then be usedas a basis for evaluating the change in thedistribution of target animals and communities,and the relation to the naturalvariability of the local ecosystem.
\n\t\t\tThe baseline, impact assessment and monitoring studiescarried out in relation to the Horns Rev 2 (Leonhard, 2006; Skov& Thomsen, 2006; Skov et al., 2008) and Anholt (Møhlenberg, 2009; Skov et al., 2009) projects highlighted the benefits of addingmodel data to the results from traditional surveys. The baseline conditions are used as a yardstick to evaluate the permanent changes in benthic habitats following establishment of the wind farms, and temporal effects related to earth works.The merits of usingcombined hydrodynamic, sediment andbiological models as a basis for estimationof environmental impacts can besummarised as:
\n\t\t\tEstimation of the realistic scale ofimpacts;
Identification of hydrographic and geomorphologicstructures and habitatsand estimation of their variability;
Increase of power of sampled data byprovision of physio-chemical data;
Improvement of understanding ofthe local dynamics of project siteand hence interpretation of changes - especially in relation to regional scaleevents;
Evaluation of the similarity of referenceand impact sites, incl. re-assessmentof the location of the referenceareas;
Evaluation of the extent of the monitoringdesign in relation to the (modelled)level of impact in monitoredareas.
The model design applied for the Horns Rev 2 and Anholt offshore wind farms is based on four model elements:
\n\t\t\tA regional and local hydrodynamic model;
An ecological model;
A deterministic filter-feeder model;
A habitat suitability model.
Several numerical 3D flow models have been established within the MIKE modelling framework covering the North Sea and Kattegat. Each of these models has individual strengths. With the purpose of water quality modelling, the so-called BANSAI model (DHI, 2006) was chosen as it has been running operationally since 2001. The model provides input data with regard to the flow field and water quality, and consists of two parts:
\n\t\t\t\tA hydrodynamic module for calculating the evolution in water levels, currents, salinity, and water temperature.
An ecological module that calculates the spreading of nutrients, the primary production, the biomass, and other ecological parameters.
The main objective of this integrated model system is to calculate the environmental status in the area of the wind farm sites. This includes source apportioning, transport, dispersion, transformation and removal in the coastal and open sea marine waters of nutrients inputs to the North and Baltic Seas. Originally the BANSAI modelwas created in a collaboration between the Swedish Meteorological and Hydrological Institute (SMHI, Sweden), Finnish Institute of Marine Research (FIMR) and DHI.

The model is using DHI’s 3-dimensional model system MIKE3 Classic, which is a fully three-dimensional, non-hydrostatic, primitive equation model (Rasmussen, 1991). It is based on the Reynolds-averaged Navier-Stokes equations and the conservation of mass, salinity and temperature. The prognostic variables are fluid pressure, the three velocity components and the two scalar quantities salt and temperature. In the waters nearest Denmark (the eastern part of the North Sea, Skagerrak, Kattegat, the Belts and the western Baltic) a 3 nautical miles grid is used while a 9 nautical miles grid is used in the North Sea and in the eastern Baltic Sea. The local model applied has this resolution in the outer mesh but by use of the nesting technique this is downscaled by a factor 9 to a resolution of app. 600 m in the area of interest where the wind mills are located. The distance between the wind mills is 600 m – 700 m which means that there will be approximately one wind turbine in each cell in the model area.
\n\t\t\t\tThe model represents the water column with a 2m resolution. The model is operational and based on:
\n\t\t\t\tMeteorology;
Tide, salinity-, temperature and nutrients on the edge of the Atlantic (tide from tidal constituents, salinity and temperature from monthly climatology (ICES), nutrients from climatology supplied with national monitoring data from Denmark and Germany;
Runoff and nutrient loadings from land (runoff from monthly climatology from HELCOM, OSPAR, national monitoring data) and nutrient loadings from climatology supplied with national monitoring data.
The model was first calibrated based on measurements from the year 2000 and has been continuously improved since then. The representation of salinity in the Belts is extremely important for ecological modelling in the Kattegat, whereas the representation of currents is the key to obtain correct ecological conditions in the eastern part of the North Sea.
\n\t\t\tThe ecological model consists of an eutrophication model describing the pelagic system with 13 state variables, and seven state variables describing the exchangeable Nitrogen and Phosphorous pools in the sediment (Rasmussen et al., 2009). The pelagic system includes phytoplankton, described in terms of their concentration of carbon (C), nitrogen (N) and phosphorus (P), chlorophyll-a, zooplankton, detritus (C, N & P), inorganic nutrients (dissolved inorganic nitrogen—DIN & PO4–P), total N and P nutrients (including dissolved organic N and P compounds) and dissolved Oxygen (DO). Due to the depth in the wind farm development areas benthic vegetation (i.e. macroalgae) is poorly developed or not existing, and accordingly benthic vegetation is not included in the model. In addition to state variables a large suite of derived variables such as water transparency and secchi depth is modelled and stored during the modelling process. Benthic organisms are not modelled explicitly, but are included as a forcing in the water quality model. Filter-feeding bivalves constitute on average 93% of the entire biomass of benthic invertebrates in the areas, and their filtering activity can exert a significant grazing loss on phytoplankton. Their effect is included in the model by imposing a filtration loss on phytoplankton and detritus in the near bed model layer according to the filtration capacity calculated from length distribution and total biomass of the different species. Because bivalves are not included as a state-variable they do not participate directly in nutrient cycling and accordingly, 50% of filtered algae (C,N,P) are returned as inorganic solutes to the near-bed layer and 50% are entered into the detritus pool subject to sedimentation and remineralisation. Figure 2 shows the state variables and processes for carbon (C) for the pelagic system.
\n\t\t\t\tThe ecological model was built using the generic equation solver ECOLab that functions as a module in the MIKE 3 simulation software, and ECOLab is linked to the advection-dispersion term of the hydrodynamic flow model, enabling transport mechanisms based on advection-dispersion to be seamlessly integrated into the ECO Lab simulation.
\n\t\t\t\tForcings and boundary conditions of the water quality model follows the line of the forcings and boundaries of the hydrodynamic model, but in addition values for all pelagic state variables at boundaries (Øresund, Southern Kattegat and north of Læsø) and nutrient concentrations in freshwater loads (monthly basis) in addition to atmospheric loads are included. Boundary values are forced with water quality data extracted from the BANSAI model.

\n\t\t\t\tThe growth functions described above relate to individual bivalves surrounded by food at constant concentrations. In nature, filter-feeding bivalves aggregate in dense assemblages if current speeds are high, e.g. in tidal areas such as in the Wadden Sea. In low-current environments plankton algae removed by filtration are only slowly replenished and such environments cannot sustain dense populations. Therefore, the growth functions need to be supplemented by an equation that describes the replenishment of food. In

\n\t\t\t\tControlled experiments of the effects of current speed on growth have only been carried out on oysters, which showed an increase until an optimal current speed of 15 cm s-1, after which the growth started decreasing. Other bivalve species such as blue mussels increase growth in the field with increasing current speed and wind-induced turbulence until a plateau. This is generally interpreted as a consequence of increasing food availability. Mussels which are settled on substrate like cliffs, stones and foundations may survive and grow in even very energy rich environments (e.g. in current speeds> 60-80 cm s-1), while blue mussels on sandy sediments are unable to establish long-living populations at current speeds exceeding 40-50 cm s-1, probably as a result of erosion.

Extended periods with low oxygen concentration can reduce growth and increase mortality in benthic invertebrates including filter-feeders. The model time series of benthic productivity provided a solid basis for the assessment of the importance of the wind farm area to waterbirds. Both baselineand historic survey data unambiguously point at the fact that the waterbirds do not use the wind farm and associated areas with lower carrying capacity for filter-feeding bivalves to any great extent (Figure 6). The areas of high carrying capacity for mussel growth (> 0.15), which matchexactly the most sensitive areas to the waterbirds in the Central Kattegat are located at a minimum distance of 8 km from the wind farm site.

Suitability functions compare the distribution of razor clams and trough shells in the multivariate oceanographic space encompassed by the recorded presence data with the multivariate space of the whole set of cells in the modelled area (Hirzel, 2001). On the basis of differences in the bivalve and the global ‘space’ with respect to their mean and variances, marginality of bivalve records was identified by differences to the global mean and specialisation by a lower species variance than global variance. Thus, for large geographical areas like the part of the North Sea studied here, ENFA approaches Hutchinson’s concept of ecological niche, defined as a hyper-volume in the multi-dimensional space of ecological variables within which a species can maintain a viable population (Hutchinson, 1957).
\n\t\t\t\tTo take account of multi-colinearity and interactions among eco-geographical factors, indices of marginality and specialisation were estimated by factor analysis; the first component being the marginality factor passing through the centroid of all bivalve presence records and the centroid of all background cells in the study area, and the index of marginality measuring the orthogonal distance between the two centroids. Several specialisation factors were successively extracted from the n-1 residual dimensions, ensuring their orthogonality to the marginality factor while maximising the ratio between the residual variance of the background data and the variances of the bivalve occurrences. A high specialisation indicates restricted habitat usage compared to the range of conditions measured in the studied part of Horns Rev. A habitat suitability index was computed on the basis of the marginality factors and the first three specialisation factors, as a high proportion of the total variance was explained by the first few factors, by comparison to a broken-stick distribution. The habitat suitability algorithm allocated values to all grid cells in the study area, which were proportional to the distance between their position and the position of the species optimum in factorial space.

Marginality coefficients showed that razor clams were (positively) linked to water depth, areas with relatively flat terrain and the carrying capacity index, while trough shells showed strong links to median grain size (negative coefficient) and the carrying capacity index. These scores can easily be interpreted on the basis of the plotted presence/absence data, which indicate that razor clams mainly use offshore areas and are found commonly around and on Horns Rev, whereas trough shells are mainly found in the eastern-most, near-coastal areas. The marginality and specialization scores lead to habitat suitability scores ranging from 0-100, the upper 33 reflecting suitable habitat (Figures 8, 9). The pixels indicating high habitat suitability for razor clams mainly lie within a coherent zone corresponding to the Horns Rev and moderate slope areas to the northwest and southeast, including the wind farm areas on Horns Rev (Figure 8). The pixels indicating high habitat suitability for trough shells (Figure 9) are confined to the area of fine sediments and high carrying capacity values in the south-eastern and eastern-most part. In most years, the wind farms areas have low suitability for trough shells, and intermediate suitability is only estimated for the Horns Rev 1 wind farm on the eastern part of Horns Rev.

\n\t\tIn the human body, the skin is the outermost and largest organ with three basic functions, i.e. protecting from various stresses and hazards, regulating some physiological aspects, and sensing any conditional changes in the environment. While conducting its functions, the skin works with other internal systems and forms a rigid network with nervous, immune, and endocrine systems [1]. On the other hand, the skin also interacts with the environment around [2] such as microorganisms living on the skin surface [3] to maintain the function of the human body. Any imbalances between these factors often lead to various multifactorial skin disorders [4]. Based on their main cause, skin disorders can be classified into six groups including tumor and cancer, trauma, pigmentation disorders, microbial (viral, bacterial, fungal, and parasitic) infections, rashes, and miscellaneous conditions [5]. Therefore, treatment of skin disorders requires systematic attention in the medical field.
The treatment of skin disorders is usually conducted in topical, systematic, or combinatorial modes using bioactive compounds to relieve any formed defects in nervous, immune, and endocrine systems. Various bioactive compounds have been evaluated through a long clinical assay [6]. In the development process of these compounds, many parameters should be considered such as economical aspect, bioavailability, stability, toxicity, and metabolism of drug molecules in both topical and systematic therapies [7].
Chalcone is a class of organic compounds with 1,3-diaryl-2-propen-1-one (Figure 1) as the backbone structure that is obtained either through an isolation and purification process from natural samples [8], semisynthesis process from existing natural products [9], or total synthesis process [10]. Chalcone is considered as privileged scaffold since it exhibits broad biological activities [10]. Many pieces of study have used chalcone as a lead compound to find more potent drugs for diseases due to the ease and convenience in synthesizing and functionalizing chalcone structure. In treating skin disorders, the efficacy of chalcone derivatives is related to several biological activities including antioxidant [11], anti-inflammation [12], immunomodulation [13], anti-angiogenesis [14], antimicrobial [15], and enzyme modulator [16]. The usage of chalcone derivatives in dermatology has been developed over the past several years to obtain the most active ingredient in either for maintaining skin health or treating skin disorders. Licochalcone A, isoliquiritigenin, and xanthohumol are examples of well-known chalcone derivatives with low toxicity and side effects on treating skin disorders and disease. In this chapter, these applications are described and discussed to provide a broad and comprehensive perspective regarding the role of chalcones in the dermatology field.
Backbone structure of chalcones.
The ultraviolet (UV) light is the electromagnetic radiation with a wavelength range from 200 to 400 nm. In general, based on the wavelength range, UV radiation is divided into four regions including UVC, UVB, UVA2, and UVA1 at 200–280, 280–320, 320–340, and 340–400 nm, respectively. This hazardous radiation causes various acute and chronic negative effects on the human skin. Clinical manifestations that occur usually depend on the wavelength and intensity of UV radiation, part of the body exposed, and type of skin (based on Fitzpatrick’s classification of skin). Acute effects of UV radiations, in general, involve various forms of inflammations such as erythema, local immunosuppression, phototanning, and epidermis thickening [17]. Mechanistically, acute effects are initiated by suitable interactions between several chromophores (either on the epidermis or dermis layers) and UV irradiation [18]. These interactions trigger the structural changes that modulate various biochemical and immunological processes [19] such as releasing several pro-inflammatory cytokines, damaging various cell biomolecules [20], generating various reactive oxygen species (ROS) [21], and producing several inflammatory mediators such as prostaglandins, histamine, and leukotrienes [22, 23]. Meanwhile, chronic effects usually lead to photoadaptation or photoprotection effects through the formation of photoaging, immunosuppression, and photocarcinogenesis [17].
Chalcone has been found as one of the bioactive compounds that is able to reduce negative effects from UV radiation such as hesperidin methyl chalcone, licochalcone A, etc. Chalcones as antioxidant and anti-inflammatory agents are used in topical or systemic methods, while the sunscreen agent is used in topical applications. In general, the sunscreen activity of chalcones is generated by high UV absorbance on the UVB-UVA region [24], which corresponds to n → π* and π → π* electronic transition from HOMO to LUMO energy levels in chalcones’ conjugated electronic system. The absorbance region of chalcones can be shifted by introducing electron-donor substituents (bathochromic shift) or electron-withdrawing substituents (hypsochromic shift) in both of the chalcone aromatic rings. However, the effects of these groups are more significant if present in A ring compared to B ring [25].
The main problem in using chalcone derivatives as sunscreen active compound is relatively poor photostability and transformations into various by-products [26, 27, 28]. Several reports have investigated the utilization of chalcone derivatives as sunscreen active compounds and also prepared sunscreen formulations with high compatibility on the skin. However, the photostability of chalcone derivatives used has not been evaluated yet [25, 26].

Similar to their flavanone analog, hesperidin [34], hesperidin methyl chalcone, and hydrolyzed methylhesperidin compounds as semisynthetic products also exhibit high UV protective effect. Topical and systematic administration of hesperidin methyl chalcone in hairless mice inhibited UVB-induced oxidative stress by reducing free radicals and ROS, enhanced endogenous antioxidant systems, and inhibited inflammation by reducing the production of cytokines [35]. The hydrolysis product of methylhesperidin, 4′,6′-dihydroxy-3,4,2′-trimethoxychalcone and 4′-hydroxy-3,4,2′,6′-tetramethoxychalcone compounds, also induced cytoprotective gene expression and reduced oxidative stress by promoting Nrf2 nuclear translocation and antioxidant response element (ARE) luciferase activity in UVB-irradiated keratinocytes [36].

Melanogenesis is a complex process of production and distribution of melanin by melanocytes. In this process, melanins (in form of eumelanin, pheomelanin, or trichochrome) are synthesized and stored in melanosomes (an organelle in melanocytes) and then transported to nearby keratinocytes to act as photoprotector in the skin and lead to chronic pigmentations [39]. The synthesis of melanin involves several reaction steps and is catalyzed by phenylalanine hydroxylase (PAH), tyrosinase (TYR), tyrosine hydroxylase isoform I (THI), tyrosinase-related protein 1 (TRYP1), and tyrosinase-related protein 2 (TRYP2) [40] enzymes. The rate-determining step of this process is hydroxylation of L-tyrosine to L-dopaquinone catalyzed by tyrosinase enzyme [41]. The final products of this process include black-brown eumelanin, yellow-reddish brown pheomelanin, and trichochrome [42].
There are many internal (endocrine, immune, inflammatory, and central nervous systems) and external (ultraviolet radiation and drugs) factors that affect the melanogenesis process [41]. Any disruptions from these factors will cause different types of pigmentation disorders including various skin conditions with strange melanocyte density, melanin concentration, or both that change pigmentations in the skin [43]. In general, pigmentation disorders can be divided into two main groups including hyperpigmentation and hypopigmentation that related to the amount of changes of normal melanin pigmentation, respectively. Both of these groups include several diseases with different clinical manifestations, but the most common forms of these are melasma (hyperpigmentations) and vitiligo (hypopigmentations) [42, 43].
Melasma is a multifactorial skin disease indicated by the presence of symmetrical hyperpigmented area in a certain part of the face including centrofacial, malar, and mandibular parts. This disease is affected by UV, visible, and infrared exposure, by inducing reactive oxygen species and promoting melanogenesis [44], hormonal conditions [45], and genetics [46]. Chalcone derivatives are extensively used in medical therapy as photoprotective described in Sections 2.1 and 2.2, and hypopigmenting agents. Various studies showed that either natural or synthetic chalcones exhibited strong activity in inhibiting cellular tyrosinase and reducing cellular melanin formation [47]. However, the action mode of chalcone derivatives in this study is unknown since most of these studies used mushroom tyrosinase that is different from human tyrosinase [48]. Other studies conducted by Kim and coworkers showed similar results with additional parameters. In these studies, chalcones containing cyclohexylmethoxy group not only attenuated cellular melanin production and tyrosinase activity, but also reduced the expressions of several melanogenesis-related genes (transcriptional activity of tyrosinase and microphthalmia-associated transcription factor/MITF) and proteins (TRP1, TRP2, and MITF) [49, 50].
Table 1 shows several in vitro assays that use Murine B16 melanoma cell lines (B16F10), melanoma cells, and human melanocyte (G361) cells in determining chalcone derivatives’ efficacy for hypopigmenting agent. In vivo studies also showed that licochalcone A and isoliquiritigenin-containing licorice extract cream can improve melasma and increase skin brightness [51]. Loading the licorice extract into solid lipid nanoparticles has been formulated and applied in a clinical trial for melisma [52].

Vitiligo is an acute skin disease characterized by unilaterally distributed depigmented areas in the skin due to the systematic degradation of melanocytes [56]. The mechanism triggering this disease has not completely elucidated yet; however, there are several theories proposed to explain it including genetics (mutations of certain genes cause autoimmunity) [57], autoimmunity (instable melanocytes induce immune system activation), oxidative stress (endogenous and exogenous stress-induced reactive oxygen species that cause internal damage of melanocytes) [58], melanocyte growth and defective melanocyte adhesion (repeated pressure and friction cause detachment of melanocytes to surrounding structures) [59], viral infections (certain viruses cause vitiligo) [60], and neural mechanism (neuropeptides elevate in vitiligo lesions) [61].
Improvement of skin appearance in vitiligo can be approached either by pigmentation or depigmentation. Chalcone derivatives are able to act as hypopigmenting (as described in Section 4.1) and hyperpigmenting agents. Several chalcone derivatives acted as hyperpigmenting agent by activating tyrosinase enzyme and elevating melanin production (Table 2). The presence of certain electron-withdrawing substituents such as halogen and trifluoromethyl substituents on the chalcone structure exhibited a significant effect in activating tyrosinase [62]. Chalcones also showed strong absorption in UV regions with low toxicity and have been formulated as broad-spectrum sunscreen protecting the affected skin from UV radiations and photofilter used in narrow-band UVB (NB-UVB) phototherapy [63].

Acne vulgaris, one of multifactorial acute inflammatory diseases, affects the pilosebaceous unit (hair follicles) in the skin. This disease is commonly found in the area with high-density pilosebaceous units (face, neck, upper chest, shoulders, and back) and characterized by the presence of seborrhea (excessive grease production), noninflammatory lesions (open comedones/blackhead and closed comedones/whitehead), inflammatory lesions (papules and pustules), and various degrees of scarring [89].

Several chalcone derivatives exhibited anti-acne activity with mainly as antibacterial and anti-inflammatory agents. Licochalcone A as a potent anti-inflammatory agent [31] has been studied and combined with other active compounds in several anti-acne formulations. Skin care formulation containing licochalcone A, L-carnitine, and 1,2-decanediol can reduce pustule lesions, popular lesions, total lesions, and sebum levels [92]. A combination of 0.1% adapalene gel and moisturizer containing licochalcone A, L-carnitine, and 1,2-decanediol also showed synergistic effect in reducing inflammatory lesions without interfering the efficacy of each active ingredient [93]. Another combination between 0.1% adapalene gel and moisturizer containing licochalcone A, glycolic acid, salicylic acid, and gluconolactone even showed better results than monotherapy using only adapalene [94]. Antibacterial effects against
Seborrheic dermatitis is a skin illness mainly influencing skin area with high amounts of sebaceous glands (such as the face, scalp, central chest, and anogenital) and characterized with the presence of erythematous patches with superficial scaling. The mildest form of seborrheic dermatitis that only affects the scalp area with no overt inflammation is usually considered as dandruff or pityriasis capitis. In general, seborrheic dermatitis affects adolescents during puberty to adulthood; however, another form of this disease is considered as infantile seborrheic dermatitis affecting babies and young children [102].
The main mechanism triggering this disease remains unknown; however, it is probably caused by the imbalance of three factors: sebum oversecretion by the sebaceous gland that is affected by certain hormonal [103] and environmental [104] conditions, sebum metabolism by

Psoriasis is a multifactorial skin disease characterized by the presence of red, dry, itchy, and scaly plaques due to chronic hyperproliferative keratinocyte cells and inflammatory cell infiltration [111]. Pathogenesis of this disease includes several steps triggered by environmental [111] and genetic [112] factors: (1) secretion of TNF-α by keratinocyte cells triggered by certain environmental and genetic factors, (2) TNF-α-induced dendritic cell activation and production of IL-23, (3) IL-23-induced T-helper 17 (Th17) cell differentiation, and secretion of IL-17A that cause hyperproliferation of keratinocyte cells [113]. Moreover, IL-17A also synergizes with IL-36 secreted by keratinocytes to amplify and support chronic characteristic of this disease [114]. Several molecular targets have been identified with drug candidates being tested in clinical stages such as cytokines (TNF-α, IL-17A, IL-17E, IL-17F, and IL-23 p19 subunit), phosphodiesterase 4, A3 adenosine receptor, and Janus kinases (JAK1-JAK3) signaling pathways [115].
Recent studies report that chalcones are efficient in improving some psoriasis models. Intraperitoneal injection of isoliquiritigenin compound remedied mouse with psoriasis-like skin predisposition and decreased several pro-inflammatory cytokines (IL-6 and IL-8) as well by inhibition of NF-κB on in vitro and in vivo assays [116]. Atopic dermatitis (atopic eczema) is a complex inflammatory skin disease characterized by the presence of skin dryness with intense itching sensation, younger/early-onset, and atopy [121]. There are three main factors that contribute to pathogenesis of atopic dermatitis, i.e., (1) genetic and immune factors on the defect of physical and chemical barrier in the skin [122], (2) hyperactive immune cells due to a biological response (mutation on caspase recruitment domain family member 11/CARD11 gene) because of the incoming pollutants and pathogens [123], and (3) environmental factors that contribute in triggering and enhancing severities of atopic dermatitis process [124].
Chalcone-containing formulations have been prepared and evaluated in treating atopic dermatitis. These chalcone derivatives were combined with other active ingredients, and it was found that they acted as anti-inflammatory and antioxidative agents. It was also reported that licochalcone A-containing moisturizer showed slightly lower effect than hydrocortisone lotion in treating mild-to-moderate atopic dermatitis in children [125]. Other compounds such as isoliquiritigenin and 3′-isopentenyl-2,2′,4,4′-tetrahydroxy-6′-methoxychalcone (ITC) showed similar effects on atopic dermatitis skin. Evaluation of isoliquiritigenin in relieving atopic dermatitis-like lesion in mice caused a significant improvement in skin condition and attenuated several pro-inflammatory parameters such as Immunoglobulin E (IgE), Th2 cytokine upregulation, TNF-α, IL-6, and IL-4 [129]. Rosacea is a multifactorial chronic inflammatory skin disorder characterized by the presence of persistent or periodical redness and several kinds of changing in phymatous in the central facial skin (cheeks, chin, nose, and central part of forehead) [131]. Based on the appearance of certain major phenotypes, rosacea is divided into erythematotelangiectatic rosacea, papulopustular rosacea, glandular/hyperplastic rosacea, ocular rosacea, and other special forms such as rosacea conglobata, rosacea fulminans, gram-negative rosacea, steroid-induced rosacea, granulomatous (lupoid) rosacea, Morbihan’s disease, and rosacea in children [132]. Pathogenesis of rosacea has not been fully comprehended yet, but there are several factors that involve including (1) genetics, Haber’s syndrome with rosacea as one of clinical feature can be inherited; (2) environment, certain environmental factors can trigger rosacea including extreme air temperature, sudden temperature changes, food (caffeine, alcohol, hot and spicy food), sunlight (UV and IR radiation), etc.; (3) overproduction of antimicrobial peptides (AMPs) by congenital immune system such as LL-37; (4) ROS-induced inflammation produced by adaptive immune cells; (5) overexpression of toll-like receptors (TLRs); (6)
Hesperidin methyl chalcone, licochalcone A, and tetracarboxymethyl naringenin chalcone exhibit significant activity in improving the skin with rosacea. Clinical studies showed that hesperidin methyl chalcone-containing topical formulations could improve infected skin condition by decreasing the proportion of dilated vessels, total vessel area, and IL8 productions. These formulations showed a complementary effect between each active ingredient in relieving inflammation and reducing the redness [134]. Another study reported that hesperidin methyl chalcone had anti-inflammatory and anti-analgesic activities which are two targets in treating rosacea by inhibiting transient receptor potential vanilloid type 1 (TRPV1), oxidative stress, TNF-α, interleukin (IL) production (IL-1β, IL-6, and IL-10), and NF-κB activity [135].

Most of the skin diseases involve any defects in the skin due to its interaction with UV irradiation, pollutant, and/or other internal and external factors. Because of that, several skin diseases such as pigmentation disorders (melasma and vitiligo), cutaneous leishmaniasis, rashes (acne vulgaris, seborrheic dermatitis and dandruff, psoriasis, and atopic dermatitis), and rosacea have been reported and investigated for better medical treatment. Chalcones, a group of privileged molecules with 1,3-diaryl-2-propen-1-one backbone, exhibit high efficacy in treating those skin diseases. These efficacies are strongly related to the good activity of chalcones as antioxidant, anti-inflammation and immunomodulation, anti-angiogenesis, antimicrobial agents, as well as their ability in modulating various enzymes. Even though chalcones are really potential to be used as a photoprotective agent, the utilization of designed synthetic chalcones is still limited due to their low photostability and medium to high toxicity and also because of unknown protein targets of those skin diseases as for today reports. Development in these fields is still required to obtain more potent drugs with excellent biocompatibility and other desired properties.

The causative agent, CPV‐2, was first identified in the late 1970s. CPV is a nonenveloped, linear, single‐stranded DNA virus with a genome of approximately 5 kb, and it belongs to the genus Parvovirus, together with feline panleukopenia virus, mink enteritis virus, raccoon parvovirus, and porcine parvovirus. An antigenic variant, CPV‐2a, identified within a few years after the emergence of CPV‐2, and another variant, CPV‐2b, began appearing in the canine population in 1984. In 2000, a novel antigenic variant, CPV‐2c, was first detected in Italy. Among the non‐infectious causes, trauma during pregnancy, genetic characteristics of the animal, deficit nutrition, thyroid dysfunction, maternal problems and hormonal disorders are found. The majority of the cases are in relation to infectious diseases, one should consider viral, bacterial, fungal and protozoal, which can interfere directly or indirectly in the foetal development. The progression of foetal development may be affected by the direct action of the microorganisms to overcome the placenta, but they are also able to affect pregnancy and release placental toxins by inflammatory processes and, may still cause maternal pathologies, which entail problems such as hyperthermia, hypoxia and endotoxemia, which can result in abortion. Several diseases can trigger pregnancy loss in dogs. This action can be direct by microorganisms, as well as indirectly triggering other problems that lead to abortion. Biochemistry examines macromolecules - proteins, nucleic acids, carbohydrates, and lipids – and their building blocks, structures, functions, and interactions. Much of biochemistry is devoted to enzymes, proteins that catalyze chemical reactions, enzyme structures, mechanisms of action and their roles within cells. Biochemistry also studies small signaling molecules, coenzymes, inhibitors, vitamins, and hormones, which play roles in life processes. Biochemical experimentation, besides coopting classical chemistry methods, e.g., chromatography, adopted new techniques, e.g., X-ray diffraction, electron microscopy, NMR, radioisotopes, and developed sophisticated microbial genetic tools, e.g., auxotroph mutants and their revertants, fermentation, etc. More recently, biochemistry embraced the ‘big data’ omics systems. Initial biochemical studies have been exclusively analytic: dissecting, purifying, and examining individual components of a biological system; in the apt words of Efraim Racker (1913 –1991), “Don’t waste clean thinking on dirty enzymes.” Today, however, biochemistry is becoming more agglomerative and comprehensive, setting out to integrate and describe entirely particular biological systems. Travieso-González received his MSc degree in Telecommunication Engineering at Polytechnic University of Catalonia (UPC), Spain in 1997, and his Ph.D. degree in 2002 at the University of Las Palmas de Gran Canaria (ULPGC-Spain). He is a full professor of signal processing and pattern recognition and is head of the Signals and Communications Department at ULPGC, teaching from 2001 on subjects on signal processing and learning theory. His research lines are biometrics, biomedical signals and images, data mining, classification system, signal and image processing, machine learning, and environmental intelligence. He has researched in 52 international and Spanish research projects, some of them as head researcher. He is co-author of 4 books, co-editor of 27 proceedings books, guest editor for 8 JCR-ISI international journals, and up to 24 book chapters. He has over 450 papers published in international journals and conferences (81 of them indexed on JCR – ISI - Web of Science). He has published seven patents in the Spanish Patent and Trademark Office. He has been a supervisor on 8 Ph.D. theses (11 more are under supervision), and 130 master theses. He is the founder of The IEEE IWOBI conference series and the president of its Steering Committee, as well as the founder of both the InnoEducaTIC and APPIS conference series. He is an evaluator of project proposals for the European Union (H2020), Medical Research Council (MRC, UK), Spanish Government (ANECA, Spain), Research National Agency (ANR, France), DAAD (Germany), Argentinian Government, and the Colombian Institutions. He has been a reviewer in different indexed international journals (<70) and conferences (<250) since 2001.
