Utilization of induced pluripotent stem cells for transplantation, treatment, and study of various liver disorders. Specific project includes generating induced pluripotent stem cells from human hepatocytes that have genetic disorders such as Crigler-Najjar Syndrome, Ornithine Transcarbamylase Deficiency, and Carbamoyl Phosphate Synthetase-I Deficiency. Once a fully reprogrammed iPS cell line is established in vitro differentiation techniques will be used to generate iPS-derived hepatocytes. These iPS-derived hepatocytes will then be transplanted into liver repopulation mouse models with the goal of generating a disease phenotype in the mouse that is human cell based.
